A diferente sensibilidade para o gosto amargo do PROP (6-n-propil-tiouracilo) está associada a fatores genéticos, mas a influência do meio oral, nomeadamente da composição salivar, não é de excluir. Neste trabalho compararam-se as características salivares de indivíduos com diferentes sensibilidades para o PROP. Foi avaliada a secreção salivar, atividade enzimática de α-amilase, pelo método do DNS e expressão de α-amílase e CA-VI, através SDS PAGE e Western blot. Não se observaram diferenças na taxa de secreção salivar, entre os grupos de sensibilidade diferente. Para a concentração proteica, as mulheres “super sensíveis” apresentam valores superiores. No que diz respeito a proteínas salivares, a atividade enzimática da α-amilase apresentou-se aumentada em mulheres “super sensíveis”. Para a CA VI, observou-se um aumento de expressão apenas em homens “super sensíveis”. Estes resultados sugerem um envolvimento das proteínas estudadas na sensibilidade gustativa, a qual difere entre géneros; ABSTRACT: The different sensitivity to the bitter taste of PROP (6-n-propylthiouracil) compound is associated with genetic factors however, the influence of the oral environment, in particular the salivary composition, cannot be excluded. In this study we compared the characteristics of saliva from individuals with different taste sensitivities to PROP. The salivary secretion, enzyme activity of α-amylase was evaluated using DNS method. α-amylase and CA-VI protein expression was evaluated using SDS PAGE and Western blot. No differences in the rate of saliva secretion, among groups of different sensitivity were observed. Concerning protein concentration, women supertasters presented higher values. With respect to the salivary proteins, the enzymatic activity of α-amylase appeared increased in women supertasters. For CA-VI there was an increase in expression only in men supertasters comparatively to the other taste sensitivity groups. These results suggest an involvement of proteins studied in taste sensitivity, and a different effect between genders.
