Estudos de neuroimagem com ressonância magnética (RM) têm sido muito valiosos na caracterização de lesões cerebrais na doença de Machado Joseph (MJD). A zona mais atingida por esta patologia é o cerebelo, contudo estudos recentes mostraram que outras estruturas cerebrais podem ser afetadas. O objetivo deste estudo é investigar possíveis alterações estruturais ou bioquímicas no estriado, hipocampo e córtex pré-frontal (PFC) em ratinhos transgénicos com idade precoce, com MJD através de RM 9,4T (sequência T2 e espectroscopia). Foram estudados 14 ratinhos saudáveis e 15 ratinhos com MJD. Foi efetuada medição volumétrica e espectroscopia protónica das três regiões de interesse. Pretende-se avaliar a possibilidade de haver alterações dos volumes cerebrais e variações dos valores de metabolitos neurotransmissores (GABA, glutamato e aspartato) e metabolitos relacionados com o metabolismo energético (tCr) nas 3 regiões de interesse entre os 2 grupos em estudo. Obteve-se uma correlação positiva entre o volume do hipocampo, estriado e a idade dos animais transgénicos com MJD. O GABA e o Glu estão correlacionados positivamente com a idade nas 3 regiões no grupo de ratinhos com MJD. O aspartato tende a ter valores mais elevados com o aumento da idade dos ratinhos com MJD no PFC. A creatina total correlaciona-se positivamente com a idade nas 3 regiões no grupo MJD. Concluíu-se que nesta fase precoce da doença ainda não são visíveis distúrbios neuronais nos animais portadores de MJD nas três regiões analisadas contudo são visíveis alterações do metabolismo energético através de espectroscopia protónica com a progressão da idade dos animais.
ABSTRACT - Neuroimaging studies with magnetic resonance imaging (MRI) have been extremely valuable in the characterization of cerebral lesions in Machado Joseph disease (MJD). The most afflicted area by this pathology is the cerebellum, although recent studies showed other brain structures might be affected. The goal of this study is the investigation of possible structural or biochemical alterations in the striate, hippocampus and pre-frontal cortex (PFC) in transgenic young mice with MJD, using a 9,4T MRI (T2 sequencing and spectroscopy). 14 healthy mice and 15 mice with MJD were studied. The 3 areas of interest were analyzed using volumetric measuring and 1H spectroscopy single-voxel. The purpose was the evaluation of cerebral volume fluctuations, and variations of the neurotransmitter metabolites (GABA, glutamate and aspartate) and energetic metabolism-related metabolites (total creatine), within the 3 regions of interest in both groups. A positive correlation between the hippocampus, the striate and transgenic MJD animals was obtained. GABA and Glu had a positive correlation with age in the 3 cerebral areas of mice with MJD. Within the PFC, Aspartate was increased with advancing age of the MJD mice. Total creatine correlates positively with age in the 3 studied areas on the MJD mice brains. It was determined that in an early stage of the disease, there are no visible neuronal alterations in animals with MJD on the 3 analyzed cerebral areas. However, as age increases, energy metabolism variations are found using 1H spectroscopy single-voxel on the 3 areas.
