This study describes the evaluation of extracts and fractions of Agave sisalana (sisal) on gastrointestinal nematodes and their toxic effects in goats. The in vitro anthelmintic activity of aqueous (AE), ethyl acetate (EE) extracts, flavonoids (FF) and saponins fractions (SF) obtained from sisal residue were determined through the egg hatch assay (EHA) and L3 migration inhibition (LMI). For the in vivo test, 30 animals were distributed into three groups: GI, was treated with AE (1.7 g/kg/day) for eight days; GII, was treated with levamisole; and GIII, was left untreated. The cytotoxic effects of extracts and fractions were investigated in Vero cells by MTT and trypan blue tests. The EC50 on eggs were 4.7, 0.1 and 0.05 mg/mL for AE, EE and FF, respectively. In the LMI, the greatest efficacy was observed in the SF (64.1%). In vivo evaluation, the reductions of faecal eggs counts were 50.3 (GI) and 93.6 (GII), and of the total number of L3 larvae were 80 (GI) and 85.6 (GII). The AE did not affect the count of adult parasites. The AE, EE, and SF were cytotoxic, with IC50 of 570, 180 and 13.7 mg/mL, respectively. The sisal showed ovicidal action and the analysis of the most active fraction (FF) indicates the presence of a homoisoflavonoid. The in vitro effect on larvae and Vero cells is related to the saponinas fraction. The in vivo anthelmintic treatment with AE showed moderate activity against eggs and free-living stages, with no evidence of toxicity.
Este estudo descreve a avalia??o de extratos e fra??es de Agave sisalana (sisal) sobre nematoides gastrintestinais e seus efeitos t?xicos em caprinos. As atividades ovicida e larvicida in vitro dos extratos aquoso (EA), acetato de etila (EE), fra??es flavono?dica (FF) e sapon?nica (FS) obtidos do res?duo de sisal foram avaliadas atrav?s dos ensaios de inibi??o da eclos?o de ovos e de migra??o larvar. No teste anti-helm?ntico in vivo, 30 animais foram distribu?dos em tr?s grupos: GI, tratado com EA (1,73 g/kg/dia) durante oito dias; GII, tratado com levamisol e o GIII, sem tratamento. O efeito citot?xico dessas fra??es foi investigado em c?lulas Vero pelos testes de MTT e azul de tripan. As CE50 sobre ovos foram de 4,7, 0,1 e 0,05 mg/mL para EA, EE e FF, respectivamente. No teste de migra??o larvar, maior efic?cia foi observada para FS (64,1%). Na avalia??o in vivo, os percentuais de redu??o da contagem de ovos foram de 50,3(GI) e 93,6 (GII), e de larvas L3 corresponderam a 80 (GI) e 85,6 (GII). O EA n?o interferiu no n?mero de parasitos adultos. Os EA, EE e FS foram citot?xicos, com CI50 de 570, 180 e 13,7 ?g/mL, respectivamente. O sisal possui a??o ovicida, e a an?lise da fra??o mais ativa (FF) indicou a presen?a de um homoisoflavonoide. Os efeitos in vitro sobre larvas e c?lulas Vero est?o relacionados com a fra??o sapon?nica. O tratamento anti-helm?ntico in vivo com o EA mostrou moderada atividade contra ovos e est?dios de vida livre, n?o apresentando evid?ncias de toxicidade.
