Interstitial Lung Disease in Mixed Connective Tissue Disease: A Single-Center C...

Direndra Hasmucrai, Khea; Costa, Filipa; Sopa, Inês; Khmelinskii, Nikita; Fonseca, João Eurico; Boleto, Gonçalo

BACKGROUND: Mixed connective tissue disease is a rare systemic autoimmune condition characterized by overlapping features of different connective tissue diseases and the presence of anti-U1-ribonucleoprotein antibodies. Its association with interstitial lung disease represents one of the main causes of morbidity and mortality in affected patients. OBJECTIVE: To characterize the clinical, immunological, and radi...

Costa, R. Pereira da; Bandeira, Matilde; Silvério-António, Manuel; Lopes, Ana Rita; Santos, Filipe Cunha; Pereira, Paulo J.; Raimundo, Diana Belchior

Background: Sjogren's disease (SjD) systemic affection is being increasingly recognized. The prevalence of peripheral (PNS) and central (CNS) nervous system involvement in SjD and risk factors for these manifestations have not been clearly established. Objectives: To define the clinical characteristics of SjD patients with CNS and/or PNS involvement and to find predictors of this involvement.

PORTRESS – the PORTuguese Reuma.pt registry for Sjögren’S disease

Bandeira, Matilde; Silvério-António, Manuel; Costa, Roberto Pereira da; Lopes, Ana Rita; Santos, Filipe Cunha; Pereira, Paulo J.

Aims: Sjögren's disease (SjD) is a complex disease with a wide variety of manifestations and outcomes. We recently created PORTRESS, the Portuguese SjD registry within Reuma.pt. We aim to describe this registry and characterize our national cohort. Methods: We included patients with a clinical diagnosis of SjD, registered in PORTRESS up to November 2023. Demographic, clinical, treatment, and patient-reported ou...

How useful is contrast-enhanced ultrasound in rheumatoid arthritis? A systemati...

Polido Pereira, Joaquim; António, Manuel; Henriques, Susana Oliveira; Sabido, João; Fernandes, Ricardo M.; Vieira de Sousa, Elsa Cristina

Objective The primary objective was to evaluate the additional benefits of contrast-enhanced ultrasound (CEUS) compared to unenhanced Doppler ultrasound (DUS) for the identification of active synovitis in rheumatoid arthritis (RA). Methods We searched PubMed/MEDLINE and EMBASE (February 2024) for all English-written published reports of human studies including the use of CEUS for the study of synovitis in RA. T...

Charneca, Sofia; Hernando, Ana; Almada-Correia, Inês; Polido-Pereira, Joaquim; Vieira, Adriana; Sousa, Joana; Almeida, Ana Santos; Motta, Carla

Background: The gut microbiota has been implicated in the onset and progression of Rheumatoid Arthritis (RA). It has been proposed that gut dysbiosis impairs gut barrier function, leading to alterations in mucosal integrity and immunity. This disruption allows bacterial translocation, contributing to the perpetuation of the inflammatory process. Since diet is recognised as a key environmental factor influencing...

The immune response to tumour development is frequently targeted with therapeutics but remains largely unexplored in diagnostics, despite being stronger for early-stage tumours. We present an immunodiagnostic platform to detect this. We identify a panel of amino acid residue biomarkers providing a signature of cancer-specific immune activation associated with tumour development and distinct from autoimmune and ...

Reproducibility and rigour in rheumatology

Silva, Augusto; Fonseca, João Eurico; Finzel, Stephanie

Scientific progress in any field of medicine, and rheumatology is no exception, is based on experimentation followed by the publication of accurate and reliable results. To achieve these aims, well-designed clinical studies need to be carried out, in such a way that other researchers can replicate them, as well as through a process of strict peer review of scientific journals. With the sustained observed expans...

Molecular maps of synovial cells in inflammatory arthritis using an optimized s...

In this study, we optimized the dissociation of synovial tissue biopsies for single-cell omics studies and created a single-cell atlas of human synovium in inflammatory arthritis. The optimized protocol allowed consistent isolation of highly viable cells from tiny fresh synovial biopsies, minimizing the synovial biopsy drop-out rate. The synovium scRNA-seq atlas contained over 100,000 unsorted synovial cells fr...

Purpose: To assess the relationship between adherence to the Mediterranean Diet (MD) /individual Dietary Inflammatory Index (DII) and disease activity, disease impact, and functional status in Rheumatoid Arthritis (RA) patients. Methods: RA patients followed at a hospital in Lisbon, Portugal, were recruited. DII was calculated using dietary intake data collected with a food frequency questionnaire (FFQ). Adhere...
