No. of bitstreams: 0 Previous issue date: 2000-01-01; Most of our knowledge concerning the virulence determinants of pathogenic fungi comes from the infected host, mainly from animal models and more recently from in vitro studies with cell cultures. The fungi usually present intra- and/or extracellular host-parasite interfaces, with the parasitism phenomen...

Gene expression during activation of Paracoccidioides brasiliensis conidia

Hernandez, O.; Garcia, A. M.; Almeida, Agostinho João Ramalho; Tamayo, D.; Gonzalez, A.; Restrepo, A.; McEwen, J. G.

This study focuses on gene expression during crucial biological phenomena of the dimorphic fungal human pathogen Paracoccidioides brasiliensis, the conidia-to-yeast (C-Y) transition and the conidia-to-mycelia (C-M) germination. We studied 10 genes involved in different cellular functions: oxidative stress response (alternative oxidase (AOX), superoxide dismutase (SOD), flavodoxin, conserved hypothetical protein...

A 32-KDa kilodalton hydrolase plays an important role in Paracoccidioides brasi...

One of the most crucial events during infection with the dimorphic fungus Paracoccidioides brasiliensis is adhesion to pulmonary epithelial cells, a pivotal step in the establishment of disease. In this study, we have evaluated the relevance of a 32-kDa protein, a putative adhesion member of the haloacid dehalogenase (HAD) superfamily of hydrolases, in the virulence of this fungus. Protein sequence analyses hav...

Genome size and ploidy of Paracoccidioides brasiliensis reveals a haploid DNA c...

Almeida, A. J.; Matute, D. R.; Carmona, J. A.; Martins, M.; Torres, I.; McEwen, J. G.; Restrepo, A.; Leão, Cecília; Ludovico, Paula

The aim of this study was to evaluate genome size and ploidy of the dimorphic pathogenic fungus Paracoccidioides brasiliensis. The cell cycle analysis of 10 P. brasiliensis isolates by flow cytometry (FCM) revealed a genome size ranging from 26.3+/-0.1Mb (26.9+/-0.1fg) to 35.5+/-0.2Mb (36.3+/-0.2fg) per uninucleated yeast cell. The DNA content of conidia from P. brasiliensis ATCC 60855-30.2+/-0.8Mb (30.9+/-0.8f...

New insights into the cell cycle profile of Paracoccidioides brasiliensis

Almeida, A. J.; Martins, M.; Carmona, J. A.; Cano, L. E.; Restrepo, A.; Leão, Cecília; Rodrigues, Fernando José dos Santos

The present work focuses on the analysis of cell cycle progression of Paracoccidioides brasiliensis yeast cells under diVerent environmental conditions. We optimized a Xow cytometric technique for cell cycle proWle analysis based on high resolution measurements of nuclear DNA. Exponentially growing cells in poor-deWned or rich-complex nutritional environments showed an increased percentage of daughter cells in ...

Compromiso pulmonar inaparente en la forma subagruda juvenil de la paracoccidio...

Restrepo, A.; Trujillo, M.; Gomez, I.
