Name of project/scholarship DJ-1 neuronal rescue under oxidative stress: implications for Parkinson's disease
Description Parkinson’s Disease (PD) is one of the major neurodegenerative disorders, characterized by massive loss of dopaminergic neurons, leading to severe motor impairment. DJ-1 was identified as a causative gene of familial early onset PD, later confirmed in animal models. Although the physiological functions of DJ-1 are not fully understood, several lines of evidence suggest that it is involved in protection against oxidative stress, the proposed cause of death of dopaminergic neurons in PD. The mechanism responsible for DJ-1-mediated neuronal rescue was thought to be direct scavenging of reactive oxygen species (ROS), however, recent data suggests that activation of survival and repression of stress- and apoptosis-related signaling pathways play the most significant role. Additional studies identified direct binding of DJ-1 to several proteins, modulating their activity (ASK-1, alfa-Synuclein, TH and L-DOPA Decarboxilase). By extensively studying DJ-1 functions, new perspectives will be opened for PD. As a result, future work may concentrate on the design of small molecules that may easily cross the blood-brain barrier and compensate for the loss of DJ-1 in patients, by artificially modulating the relevant signaling pathways identified.
