Photoresponsive hydrogels as a therapeutic platform for cancer treatment
| Resumo: | Targeted therapies for tumour treatment have emerged as promising approaches for inducing effective anti-tumour responses while reducing side effects in healthy tissues. However, the low bioavailability of many of the compounds used in these therapeutic strategies continues to limit their clinical application. In order to overcome these limitations, the main objective of this dissertation was to develop a new polymeric formulation in its native and acrylated form for the formation of hydrogels. The incorporation of porphyrin derivatives in these platforms was also promoted through covalent bonding, thus constituting a multifunctional system. To achieve this objective, porphyrin derivatives were prepared and subsequently functionalised with methacrylate derivative, obtaining porphyrin derivatives with different degrees of substitution. Similarly, the polymer matrix was functionalised and the conjugation between acrylated and non-acrylated polymer matrices with porphyrin derivatives was promoted, obtaining polymer matrices conjugated with porphyrins. All porphyrin derivatives and polymer conjugates prepared were subjected to structural characterisation by ¹H, ¹³C and ¹⁹F NMR and, whenever possible, by mass spectrometry and elemental analysis, thus confirming the structures obtained and the incorporation/conjugation of porphyrin into the polymer matrix. Finally, several hydrogel formulations were prepared based on the polymer matrices conjugated with porphyrin in order to evaluate the behaviour of the hydrogel and the influence of the composition on the photoactive response and properties of the material.
