Gold (I) Complexes as New Fluorescent Sensor Systems for Nucleotides

Resumo:The detection of small analytes is a fundamental study nowadays. Since nucleotides are the basic constituents of nucleic acids and are involved in many metabolic processes, the development of detection tools is essential for detecting such analytes as well as larger molecules such as DNA and some proteins composed essentially of adenine nucleotides. The main objective of this project was the synthesis and characterisation of a gold (I) complex for the development of a new fluorescent sensor for the recognition of small analytes, namely adenine-based nucleotides. The key molecule used as an adenine complementary unit, a naphthalimide ligand, was obtained with a yield of 68,9% and characterised by NMR and spectroscopic technics and the interaction studies were followed by fluorescence spectroscopy. However, some of the association constants are apparent ones, since it was not possible to reach an analyte-ligand/complex adduct formation plateau. For the complex stability, the results showed that, under certain conditions, this molecule is not as stable as expected, since complex to ligand conversion appears to occur over a short period of time. One of the most unexpected results due to the instability of this complex was the formation of gold nanoparticles. However, the results showed that the final complex can be used as a fluorescent sensor for this type of analyte, but it must be taken in consideration that all the future studies must be performed ex vivo, once the solvent mixtures must have a higher concentration of DMSO than the limit concentration used in in vivo studies. Since adenine-based nucleotides are an important group of molecules present in several biochemical processes, the detection of this type of analytes is an interesting task and the development of new methods, like fluorescence sensors, is a fundamental study to understand and improve these metabolic pathways.
