The influence of sucrose on paracetamol release from HPMC matrix at different conditions
| Resumo: | “Medicine” is defined as any substance or combination of substances presented for treating or preventing diseases in human beings or for establishing a diagnostic. Or also, throw a pharmacological, immunological or metabolic action, be able to restaur, correct or modify physiological functions (DL - 176/2006). However, for the medicine reach its therapeutic target correctly and in the intended dose, is necessary that the drug and it dosage form, in which is veiculate, be indicated for it route of administration. The interaction between drugs and the nutrients from food intake are known in many cases. Studies done previously showed that there is an interaction between dietary sugars and the matrix of hydroxypropyl methylcellulose of prolonged release tablets, and thus jeopardize the safety and efficacy of the medicine. Therefore, the aim of this study was to verify the influence of sucrose (one of the dietary sugars) in the release of paracetamol from an hydroxypropyl methylcellulose matrix. It was also tested the concentration necessary to verify this influence, also simulating the conditions in the gastrointestinal tract. The results showed a clear effect of sucrose on the stability of the tablet matrix, causing a different release of paracetamol (faster). The rate of release of paracetamol was increased as the sucrose concentration in the medium. |
