Controlo de qualidade em vinhos do Douro e Porto Utilização do método FTIR e ELISA na rotina do Instituto dos Vinhos do Douro e Porto

Resumo:During the analysis of both still and sweet wines, either for the purposes of quality control or for the attribution of denominação de origem (DO) or indicação geográfica (IG) certification, several laboratory methods are used. Among these methods, the Fourier Transform Infrared Spectroscopy (FTIR) and Enzyme-Linked Immunosorbent Assays (ELISA) are used by the Institute of Douro and Port Wines, I.P. (IVDP) during their daily sample testing, with several different purposes. During the hereby discussed internship, the student took part in the Institute’s departments where these methods are used. The process began with a general familiarization with the methods themselves, as well the procedure for the daily quality control of the results obtained. In later stages, there were attempts to improve some of the existing calibrations for the FTIR analysis equipment, and a continuation of the method validation process for the ELISA analysis technique. FTIR analysis has revealed to be a particularly fit method for obtaining significate results, both in the Douro as well as in Port wines, with the only caveat of requiring constant quality control over these results. On the other hand, the ELISA method requires greater care in the execution of the analysis itself, since it can be more sensitive to interfering substances, despite being able to detect very small amounts of the analyte being tested.
