Improvement of gene transfer mediated by Agrobacterium and transgenic plant regeneration of Portuguese table grape Dona Maria (Vitis vinifera L.) in order to overexpression of the gene encoding the stilbene synthase

Resumo:Since beginning of crops domestications that grapevine has had a huge impact in agriculture. Then which its cultivation has been high and its product have gain importance in a commercial level, worldwide. The table grape is usually suitable for consumption as fresh fruit. Characteristics like size, shape and colour of berries allow its commercialization. Dona Maria is a white table typically Portuguese that has important characteristics which becomes very attractive in market. Resveratrol is a phytoalexin with high therapeutic properties, which becomes important in many levels. It is important the study of this cultivar to improve the production of resveratrol. of resveratrol. Agrobacterium tumefaciens, used as a vector in protocols of gene transfer, have a high efficiency in the transferred of genes with various genetic construction, allowing the improvement of certain characteristics. In this work was stablished protocol of genetic transferred using embryogenic calluses of Dona Maria and Pinot Noir. It was used Agrobacterium tumefaciens as a mediator of gene transfer, with 2 different plasmids (p35SGUSINT and pHKvst). Were done 4 genetic transfer processes using selective pressure initially with 150 mg/L of kanamycin after 3 weeks of coculture. It was made an alternative process using three different methods of regeneration and allowing, after that, the comparison of their efficiency. During selective pressure step, it was possible observe embryos in different phases of development. Some of them converted in plantlets.
